Microbial community structure and dynamics of starch-fed and glucose-fed chemostats during two years of continuous operation

Min GOU,Jing ZENG,Huizhong WANG,Yueqin TANG,Toru SHIGEMATSU,Shigeru MORIMURA,Kenji KIDA

Frontiers of Environmental Science & Engineering 2016, Volume 10, Issue 2,   Pages 368-380 doi: 10.1007/s11783-015-0815-9

Abstract: The microbial community structures of two mesophilic anaerobic chemostats, one fed with glucose, the other with starch as sole carbon sources, were studied at various dilution rates (0.05–0.25 d for glucose and 0.025–0.1 d for starch) during two years continuous operation. In the glucose-fed chemostat, the aceticlastic methanogen spp. and hydrogenotrophic methanogen spp. predominated at low dilution rates, whereas spp. and the hydrogenotrophic spp. predominated together when dilution rates were greater than 0.1 d . Bacteria affiliated with the phyla Bacteroidetes, Spirochaetes, and Actinobacteria predominated at dilution rates of 0.05, 0.1, and 0.15 d , respectively, while Firmicutes predominated at higher dilution rates (0.2 and 0.25 d ). In the starch-fed chemostat, the aceticlastic and hydrogenotrophic methanogens coexisted at all dilution rates. Although bacteria belonging to only two phyla were mainly responsible for starch degradation (Spirochaetes at the dilution rate of 0.08 d and Firmicutes at other dilution rates), different bacterial genera were identified at different dilution rates. With the exception of Archaea in the glucose-fed chemostat, the band patterns revealed by denaturing gradient gel electrophoresis (DGGE) of the microbial communities in the two chemostats displayed marked changes during long-term operation at a constant dilution rate. The bacterial community changed with changes in the dilution rate, and was erratic during long-term operation in both glucose-fed and starch-fed chemostats.

Systems biomedical analysis of Schistosoma japonicum

Ze-Guang HAN MD,

Frontiers of Medicine 2010, Volume 4, Issue 2,   Pages 157-165 doi: 10.1007/s11684-010-0034-5

Abstract: Human schistosomiasis, caused mainly by three principal species including , , and , remains a major public health concern worldwide. is prevalent in southern China, being a major disease risk for 66 million people. The blood fluke has a complex life cycle for survival: as a free-living form in fresh water and as a parasite in the snail intermediate and vertebrate definitive hosts. Systems-based biomedical analyses, including genomic, transcriptomic, proteomic and metabonomic approaches, have been performed on the schistosome. These comprehensive investigations have not only characterized the genomic features but also chartered gene and protein expression profiles across genders and developmental stages. The integration of the huge information will lay a global and solid foundation for the molecular architecture of the biology, pathogenesis, and host-parasite interactions of the human blood fluke, which will facilitate the development of a new antischistosomal vaccine and drugs as well as diagnostic markers for the treatment and control of schistosomiasis.

Numerical analysis and experimental investigation of modal properties for the gearbox in wind turbine

Pengxing YI,Peng HUANG,Tielin SHI

Frontiers of Mechanical Engineering 2016, Volume 11, Issue 4,   Pages 388-402 doi: 10.1007/s11465-016-0404-z

Abstract:

Wind turbine gearbox (WTG), which functions as an accelerator, ensures the performance and service life of wind turbine systems. This paper examines the distinctive modal properties of WTGs through finite element (FE) and experimental modal analyses. The study is performed in two parts. First, a whole system model is developed to investigate the first 10 modal frequencies and mode shapes of WTG using flexible multi-body modeling techniques. Given the complex structure and operating conditions of WTG, this study applies spring elements to the model and quantifies how the bearings and gear pair interactions affect the dynamic characteristics of WTGs. Second, the FE modal results are validated through experimental modal analyses of a 1.5 WM WTG using the frequency response function method of single point excitation and multi-point response. The natural frequencies from the FE and experimental modal analyses show favorable agreement and reveal that the characteristic frequency of the studied gearbox avoids its eigen-frequency very well.
